Occurrence and Distribution of Schoenoplectus juncoides Resistant to ALS-Inhibitors in Paddy Field of Chungnam Province

초록 | The continuous use of acetolactate synthase (ALS) inhibitors has developed resistant weeds such as Monochoria vaginalis, Echinochloa crus-galli, and Schoenopplectus juncoidesin paddy field. In Korea, Monochoria korsakowiiwas first developed as a resistance to ALS inhibitor in 1988. Since then, total 13 weed species were identified as a resistance to ALS inhibitors. The herbicide resistant S.juncoides was first found in paddy field of Jeonbuk province of Korea. In order to investigate occurrence and distribution of herbicide resistant S.juncoides in paddy fields, we collected soil samples from 264 paddy fields in Chungnam province in 2017. The sampling sites were determined based on the rice cultivation area. The soil samples were put in a plastic box (L:18 cm, M:12 cm, H:10 cm) mixed with water and kept flooding at 5 cm depth. S.juncoides were tested by using 75 g a.i./ha imazosulfuron and 30 g a.i./ha pyriminobac-methyl. The number of survived S.juncoides was counted 30days after herbicide treatment. Result showed that herbicide resistant S.juncoides were found 27 sampling sites out of 264 paddy fields. The survived plants were transplanted in round pots (φ:20 cm, H:17 cm) for producing seeds. To confirm resistance study were conducted using imazaquin, penoxsulam, and flucetosulfuron in 2018. Each herbicide treated three concentrations (3000, 300 and 30 g a.i./ha). The growth-inhibitionwas checked by visual rating scale. Result showed that 14 herbicide resistant biotypes were found. Especially, one cross-resistant biotype was found. Molecular study was conducted in 2018. DNA from leaf was extracted using a DNeasy Plant Mini Kit. Gene sequences were compared between herbicide resistant and susceptible biotypes. Results showed that the substitution of amino acid in resistant biotypes was observed at Pro197 position of ALS sequence. The sequence of three resistant biotypes showed a serine codon replacing Pro197. We do not know yet the mechanism of resistance in other biotypes. Non-target site herbicide resistance should be researched such as metabolism and transport of herbicide. |
