Arenicin-1 elicits bacterial apoptotic like response Escherichia coli

초록 | Arenicin-1, a 21-mer antimicrobial peptide, has been known to exert significant broad-spectrum antimicrobial activity with membrane active mechanisms. Bacterial mechanism need to elucidate further due to work several mechanisms for cell death. In the present study, the findings suggest a novel mechanism for the antibacterial effect of arenicin-1 on Escherichia coli, namely, a bacterial apoptosis-like death (ALD). We focused on a possible mechanism in arenicin-1-treated bacterial cells that includes the accumulation of reactive oxygen species (ROS), increased intracellular calcium levels, destruction of membrane potential, bacterial caspase-like protein activation, and DNA damage. Arenicin-1 induced ALD induced arenicin-1 includes the autocleavage of LexA due to RecA expression. A mutant strain of the RecA and LexA proteins, associated with the SOS response, was used to determine the necessity for RecA/LexA proteins in arenicin-1-mediated ALD. Furthermore, superoxide, hydrogen peroxide, and hydroxyl radicals, which are typical ROS, are removed from the cells treated with arenicin-1. Hydrogen peroxide interfere with the activity of arenicin-1 in E. coli, whereas superoxide and hydroxyl radical do not affect the activity of ALD induced by arenicin-1. Also, inhibition of Fenton reaction was attenuated the feature of ALD. Taken together, arenicin-1, which is required by RecA/LexA and is interrupted by hydrogen peroxide and Fenton reaction, induces ALD response in E. coli. The novel mechanisms of arenicin-1 may be an effective antimicrobial agent with far lower propensity for inducing microbial resistance than antibiotics. |
