초록 | Cytochrome P450 proteins are abundantly present in plants and required for the catalysis of many reactions, typically in production of primary and secondary metabolites. The functions of P450s normally involve the biochemical synthesis pathways comprise terpenoids, fatty acids, lipids, alkaloid, as well biosynthesis of plant hormones, especially, the catalysis of many products for chemical defense mechanism is their outstanding role. Tomato is well-known as one of plants contains numerous CYP genes most in their genome, but most of them have unknown function. To reveal their function, a P450 in CYP736A subfamily, CYP736A72 gene, was isolated from tomato (Solanum lycopersicum cv.Micro-Tom). To study the enzyme character of CYP736A72, this gene was heterologously expressed in E. coli, and the protein obtained after induction was used for analytical assays. The initial results show that this enzyme has 7-ethoxycoumarin O-deethylation activity. To identify CYP736A72’s function in plant, the transgenic lines of tomato and tobacco (Nicotiana tabacum cv.Xanthi-NC) which over-expressed CYP736A72 gene were produced and observed. |
