First Report of a Typical Calico-Associated Isolate of Peach latent mosaic viroid from Calico Disease-Affected Peach Trees in Korea

초록 | Peach latent mosaic viroid (PLMVd), a member of the genus Pelamoviroid in the family Avsunviroidae, may be symptomless or causes yellow, chlorotic mosaics and delays in foliation, flowering, and ripening, with fruit deformation and discoloration. In June 2016, naturally infected peach trees (Prunus persica (L.) Batsch) cv. Akatsuki showing severe chlorosis of leaves, stems, and fruits were observed in Gyeongsan, South Korea, symptoms similar to the PLMVd-induced peach calico (PC) disease (e.g., Malfitano et al. 2003). Leaf tissue samples were collected individually from four symptomatic plants in the same commercial field. Presence of PLMVd was confirmed by RT-PCR using total RNAs extracted with the NucliSENS easyMAG system (bioMerieux), and specific primer pairs RF43 and RF44 (Malfitano et al. 2003). The expected PLMVd PCR fragments of ca. 350 bp were obtained and directly sequenced from all four samples. Two distinct sequences, named PC-K1 (two trees; LC311883) and PC-K2 (two trees; LC311884) were obtained, with 93 and 91% identity to PLMVd isolate PC-C40 (AJ550912), respectively. The two strains were 349 to 350 nt like PLMVd isolates with the 12 to 14 nt hairpin insertion causing PC symptoms (348 to 351 nt) (Malfitano et al. 2003), unlike the 337 to 339 nt isolates previously reported in Korea, which included the only isolate (PLMVd-Sumi, 337 nt) from a symptomatic tree (Jo et al. 2015, 2016). Rodio et al. (2006) have reported two types of insertions in PLMVd variants, with PC associated only with type 1 insertions; they also reported that insertions could both emerge and be lost during infection. PC-K2 has a type 1 insertion with a UUUU loop in the hairpin, whereas PC-K1 has a CUUU loop distinct from either type 1 or type 2 variants reported by Rodio et al. (2006). In phylogenetic trees, both PC-K1 and PC-K2 were included in a clade consisting of known PC-associated isolates and the 337-nt reference isolate (NC 003636), whereas PLMVd-Sumi and other 336 to 337 nt isolates fell into a separate clade with 95% bootstrap support. To our knowledge, this is the first report of a PLMVd isolate with the typical calico-associated hairpin insert inducing PC of peach trees in Korea; the potential emergence of PC-inducing PLMVd variants from latent isolates suggests that certification schemes for peach in Korea should include freedom from PLMVd (Rodio et al. 2006). |
