First Report of Cymbidium Chlorotic Mosaic Virus Infection in Cymbidium goeringii in South Korea.

초록 | Cymbidium is one of the most popular and commercially important cultivated orchids in the world. Cymbidium goeringii Reichenbach ill (spring orchid) grows in the wild in China, Japan, South Korea, and Southeast Asia. Among the 41 orchid-infecting viruses known to occur in Korea, Cymbidium mosaic virus (CymMV) and Odontoglossum ringspot virus (ORSV) are the most prevalent and economically important (Chung et al. 2010). In 2017, typical viral symptoms, including chlorotic mosaic and streaks, were observed on the leaves of C. goeringii in Nonsan, South Korea. Four leaf samples exhibiting symptoms were analyzed by transmission electron microscopy (TEM) of leaf dip-preparations using negative staining techniques (Horne and Wildy 1979). Typical spherical particles, with a diameter of approximately 28 nm, were observed in the leaf samples of C. goeringii plants. In the same samples, filamentous or rod-shaped particles resembling ORSV or CymMV were not observed using TEM. The presence of ORSV, CymMV, and cucumber mosaic virus (CMV) was also evaluated using respective ImmunoStrip kits (Agdia, U.S.A.) with positive and negative controls (leaves of healthy Cymbidium sp.), but none of the three targeted viruses could be detected. Subsequently, total RNA was extracted using the RNeasy Plant Mini Kit (Qiagen, Germany), and a library for transcriptome analysis of C. goeringii was prepared using the TruSeq RNA Library Preparation Kit version 2 (Illumina, U.S.A.), according to the manufacturer’s instructions. The library was used for paired-end sequencing with the Illumina HiSeq 2000 Sequencing System (Macrogen, South Korea). De novo assembly of the raw data obtained was done using the program Trinity (Haas et al. 2013). The assembled sequences were aligned with viral reference genomes by searches using BLAST for identifying any viruses infecting these orchids, and sequences corresponding to Cymbidium chlorotic mosaic virus (CyCMV; family Solemoviridae, genus Sobemovirus) were identified by a de novo transcriptome assembly. The complete genome sequence of the CyCMV isolate Kor (CyCMV-Kor) was deposited to GenBank (accession no. LC381945) after confirmation of the 5ʹ and 3ʹ end of the CyCMV genome using the SMARTer RACE kit (TAKARA Bio, U.S.A.). The presence of CyCMV in the four symptomatic leaf samples of C. goeringii was confirmed by reverse-transcription polymerase chain reaction (RT-PCR) using specific primers (forward 5ʹ-TTAATGGCGAAGCGCAAATC-3ʹ [position 3,187 to 3,260] and reverse 5ʹ-TCATCGGTTCAACGAGCTAGC-3ʹ [position 3,958 to 3,978]), which were designed based on the genome of the CyCMV isolate Cym92-20 (accession no. NC_027123.1). The RT-PCR products amplified were cloned into pGEM-T Easy vector (Promega, U.S.A.) and sequenced. The fragments amplified by PCR were 792 bp long. A BLAST search revealed that the nucleotide and amino acid sequences of the genes of the four CyCMV isolates studied, coding for coat proteins, were closely related (91.3 to 91.6% similarity) with those of Cym92-20 (Kondo et al. 2015). Since C. goeringii was imported from China, it has been cultivated in Nonsan, South Korea, for 10 years. Although it is propagated by division of pseudobulbs, it is likely that CyCMV can be spread by cutting tools to all regions of South Korea (Kondo et al. 2015). Therefore, further investigation of geographical distribution and sequence variation of CyCMV isolated from C. goeringii is necessary. To the best of our knowledge, this is the first report on the occurrence of CyCMV in Cymbidium plants in Korea. |
